Prior to securing a more prominent part as Brandon Walsh in the Richard Donner-directed film The Goonies (1985), Brolin began his career in TV movies and cameo appearances on TV series.

He was a candidate for the television series 21 Jump Street’s Tom Hanson part; Brolin and Johnny Depp were the two finalists, and the two grew close and stayed friends. In the end, Depp was cast in the part. During the first season of the show, Brolin made a cameo appearance in one episode.

The announcement that Brolin would portray Thanos in the Marvel Cinematic Universe was made in 2014. He does both voice acting and motion capture work to represent the character.

After making brief appearances as the character in Guardians of the Galaxy (2014) and Avengers: Age of Ultron (2015), Brolin returned to the role of Thanos in the back-to-back films Avengers: Infinity War (2018) and Avengers: Endgame (2019). Brolin performed as a different Thanos in the 2021 film What If…?

In April 2017, Brolin agreed to a four-picture deal with 20th Century Fox Studios to play the X-Men movie series Nathan Summers/Cable, a character from Marvel Comics. Within that arrangement, his first installment was Deadpool 2 (2018). Up until Disney acquired 20th Century Fox, he was scheduled to return to the role of X-Force in the Fox film project.
