Fatomilola was born on January 16, 1946, in Ifisin-Ekiti, situated in the Ido-Osi Local Government area of Ekiti State, Nigeria.
He is the offspring of Chief Abraham Ojo Fatomilola and Mrs. Elizabeth Fatomilola. As the son of Chief Ifa Priest, known as the Oluwo (Olu-awo, meaning Lord of the oracle), Fatomilola’s familial ties to spiritual practices influenced his portrayal of herbalist roles in Nigerian films. His mother hailed from Isare-Oge in Kwara State.

In 1967, Fatomilola joined the Olokun Theatre group, where he received guidance under the mentorship of the late Professor Ola Rotimi, a renowned dramatist and prolific playwright at the University of Ife, now Obafemi Awolowo University.
