De Matteo, of Italian heritage, was raised in a Catholic family. She pursued her education at the Loyola School before obtaining a Bachelor of Fine Arts in film production from New York University's Tisch School of the Arts. Initially aspiring to be a film director, she further honed her craft by studying acting at the HB Studio.

Her breakout role as Adriana La Cerva in The Sopranos marked the beginning of her successful career.
